Three-dimensional reconstruction of the giant mimivirus particle with an x-ray free-electron laser.

چکیده
We present a proof-of-concept three-dimensional reconstruction of the giant mimivirus particle from experimentally measured diffraction patterns from an x-ray free-electron laser. Three-dimensional imaging requires the assembly of many two-dimensional patterns into an internally consistent Fourier volume. Since each particle is randomly oriented when exposed to the x-ray pulse, relative orientations have to be retrieved from the diffraction data alone. We achieve this with a modified version of the expand, maximize and compress algorithm and validate our result using new methods.

Single-shot diffraction data from the Mimivirus particle using an X-ray free-electron laser
Free-electron lasers (FEL) hold the potential to revolutionize structural biology by producing X-ray pules short enough to outrun radiation damage, thus allowing imaging of biological samples without the limitation from radiation damage. Thus, a major part of the scientific case for the first FELs was three-dimensional (3D) reconstruction of non-crystalline biological objects. In a recent publi...

In this paper, a free-electron laser (FEL) growth rate with a laser wiggler in which plasma background is used to generate short wavelengths in x-ray regimes, has been investigated theoretically. A linearly polarized laser pulse, due to having short wiggler periods (inrange) is able to produce coherent radiations in x-ray regions and can be applied as a planar wiggler in a FEL. Phase velocity o...
متن کاملModel-independent particle species disentanglement by X-ray cross-correlation scattering
Mixtures of different particle species are often investigated using the angular averages of the scattered X-ray intensity. The number of species is deduced by singular value decomposition methods. The full disentanglement of the data into per-species contributions requires additional knowledge about the system under investigation. We propose to exploit higher-order angular X-ray intensity corre...
متن کاملMicroscopic Characterization of the Brazilian Giant Samba Virus
Prior to the discovery of the mimivirus in 2003, viruses were thought to be physically small and genetically simple. Mimivirus, with its ~750-nm particle size and its ~1.2-Mbp genome, shattered these notions and changed what it meant to be a virus. Since this discovery, the isolation and characterization of giant viruses has exploded. One of the more recently discovered giant viruses, Samba vir...
متن کاملThree-dimensional reconstruction for coherent diffraction patterns obtained by XFEL
The three-dimensional (3D) structural analysis of single particles using an X-ray free-electron laser (XFEL) is a new structural biology technique that enables observations of molecules that are difficult to crystallize, such as flexible biomolecular complexes and living tissue in the state close to physiological conditions. In order to restore the 3D structure from the diffraction patterns obt...
